This work aims to determine the effect of genotype x environment (GxE) interaction that influence blackcurrant (Ribes nigrum) fruit quality. We applied metabolomics-driven analysis on fruits from four cultivars grown in contrasting European-locations over two seasons. By integrating metabolomics and sensory analysis, we also defined specific metabolic signatures associated with consumer acceptance. Our results showed that rainfall is a crucial factor associated with accumulation of delphinidin- and cyanidin-3-O-glucoside, the two mayor blackcurrant pigments meanwhile temperature affects the main organic acid levels which can be decisive for fruit taste. Sensorial analysis showed that increases in terpenoid and acetate ester volatiles were strongly associated with higher appreciation score, while proacacipetalin, a cyanogenic-glycoside, was positively associated to bitter taste. Our results pave the way for the selection of high-quality cultivars and suitable production sites for blackcurrant cultivation.

Promoting the consumption of fruits is a key objective of nutrition policy campaigns due to their associated health benefits. Raspberries are well appreciated for their remarkable flavor and nutritional value attributable to their antioxidant properties. Consequently, one of the objectives of present-day raspberry breeding programs is to improve the fruit's sensory and nutritive characteristics. However, developing new genotypes with enhanced quality traits is a complex task due to the intricate impacts genetic and environmental factors have on these attributes, and the difficulty to phenotype them. We used a multi-platform metabolomic approach to compare flavor- and nutritional-related metabolite profiles of four raspberry cultivars ('Glen Ample', 'Schönemann', 'Tulameen' and 'Veten') grown in different European climates. Although the cultivars appear to be better adapted to high latitudes, for their content in soluble solids and acidity, multivariate statistical analyses allowed us to underscore important genotypic differences based on the profiles of important metabolites. 'Schönemann' and 'Veten' were characterized by high levels of anthocyanins and ellagitannins, respectively, 'Tulameen' by its acidity, and 'Glen Ample' for its content of sucrose and ß-ionone, two main flavor contributors. Our results confirmed the value of metabolomic-driven approaches, which may foster the development of cultivars with enhanced health properties and flavor.

Phytoplasmas are plant-pathogenic bacteria that cause a disease in Rubus species which is referred to as Rubus stunt. As phytoplasmas can be spread by vegetative propagation and latency periods of Rubus stunt can be up to one year, the use of pathogen-free Rubus propagation material in plant nurseries is important in order to stop the spread of this disease. Even though heat therapy has been commonly applied against viruses in many plants, its potential for phytoplasma eradication has been much less explored. Here, the efficacy of heat therapy with subsequent tissue culture to eliminate phytoplasmas from infected raspberry and blackberry plants is evaluated. Heat therapy was performed on 25 phytoplasma-infected raspberry and 33 infected blackberry plants, out of which 100 raspberry and 65 blackberry plants were regenerated via subsequent tissue culture. All plants were negative for the presence of phytoplasma DNA by qPCR at the end of cultivation periods of 481 to 565 days for the treated raspberry plants and 231 to 337 days for the treated blackberry plants. These results show the suitability of heat therapy combined with tissue culture as a routine tool to ensure the presence of phytoplasma-free Rubus mother plants in nurseries.

Rubus stunt is an economically important disease in the production of raspberries, blackberries, and loganberries. A fast, sensitive, and reliable diagnosis of phytoplasmas, the causal agent of the disease, is of prime importance to stop its spread by vegetative propagation and by insect vectors. Therefore, multiplex qPCR assays using TaqMan probes with different kinds of fluorophores in one reaction were developed, allowing the detection of phytoplasmas in general as well as a more specific detection of phytoplasmas belonging to group 16SrV and host DNA (either plant or insect). This assay now provides a practical tool for the screening of motherplants and monitoring the presence and distribution of phytoplasmas in Rubus plants of different geographic origins, cultivars, and cultivation systems, as well as in putative insect vectors like leafhoppers.

The increasing use of biological control agents (BCAs) against Botrytis cinerea in strawberry raises the question of whether there are any undesirable impacts of foliar applications of BCAs on nontarget microorganisms in the phyllosphere. Therefore, our objective was to investigate this issue within a field study. Strawberry plants were repeatedly sprayed with three BCAs-namely, RhizoVital 42 fl. (Bacillus amyloliquefaciens FZB42), Trianum-P (Trichoderma harzianum T22), and Naturalis (Beauveria bassiana ATCC 74040)-to suppress Botrytis cinerea infections. Microbial communities of differentially treated leaves were analyzed using plate counts and pyrosequencing and compared with the microbial community of nontreated leaves. Plate count results indicate that the applied Bacillus and Trichoderma spp. survived in the strawberry phyllosphere throughout the strawberry season. However, no significant impacts on the leaf microbiota could be detected by this culture-dependent technique. Pyrosequencing of internal transcribed spacer ribosomal RNA and 16S RNA sequences revealed a change in fungal composition and diversity at class level after the introduction of T. harzianum T22 to the phyllosphere, whereas the bacterial composition and diversity was not affected by either this Trichoderma preparation or the other two BCAs. Our results suggest that pyrosequencing represents a useful method for studying microbial interactions in the phyllosphere.

Strawberry cultivars grown at different locations in Europe showed genotype- and environment-dependent variation in total phenol and anthocyanin contents. This study focused on the compositional diversity of tannins from these cultivars using a high-throughput liquid chromatography-mass spectrometric (LC-MS) technique. Cultivars grown in Germany showed differences in the relative amounts of proanthocyanidins (PACs) and ellagitannins (ETs). Cultivars grown at three different European locations showed differences in their PAC/ET balance. 'Elsanta' grown in Switzerland had higher levels of ET-derived than PAC-derived signals compared to 'Elsanta' grown in Germany and Denmark. The trend to increased ET-derived signals was also noted for 'Clery' grown in Switzerland over Germany and was apparent for 'Korona' grown in Denmark over Germany. The altered ET/PAC balance was confirmed by conventional LC-MS analysis, which highlighted specific changes in composition rather than a general increase in ET components. These findings are discussed with respect to the environmental conditions at the different European locations.

Strawberries (Fragaria × ananassa Duch. cvs. Everest, Elsanta) were grown in a tunnel covered with two films, which were distinguished in their ultraviolet transparency, as well as under open-field conditions. One applied film was not transparent for UVB radiation, and the second film transmitted 70% of UVB radiation. During the present study, the nutritional value and quality parameters of the fruits were evaluated. Strawberries were UV-unresponsive in view of the content of ascorbic acid and sum parameters like total anthocyanins and antioxidant capacity measured with TEAC (trolox equivalent antioxidant capacity), ORAC (oxygen radical absorbance capacity) and total phenols. These parameters were mainly affected by sampling date and cultivar. However, HPLC analysis showed that individual phenolics were affected in the absence of UV radiation. The content of the anthocyanin cyanidin 3-glucoside and the flavonols quercetin 3-glucuronide and kaempferol 3-glucoside was decreased in the fruits grown under UV blocking film compared to open-field grown strawberries. By means of the UV transparent film the content of the mentioned flavonoids could be enhanced up to similar amounts like in open-field grown strawberries. All other phenolics were not consistently affected by UV radiation. This result was independent of cultivar.
